Poster Title: Method Development and Validation for the Determination of Two Protein Biomarkers by Multiplexed Online and Offline Hybrid LC-MS/MS in Human Heart Tissue

Can you explain your role at KCAS Bio?
I am a Scientist 1 at KCAS Bio in the Mass Spectrometry Bioanalysis Method Development group. In this role, I develop and optimize LC-MS/MS methods used to detect and quantify drugs, biomarkers, and other analytes in biological samples. These include many different types of modalities from small molecule biomarkers to large molecule biotherapeutics utilizing “traditional” LC-MS/MS as well as hybrid LC-MS/MS technologies. My responsibilities include sample preparation, instrument operation and troubleshooting, data analysis, and ensuring methods meet regulatory and scientific standards. Ultimately, my work supports studies that help determine how drugs are absorbed, distributed, metabolized, and eliminated in the body, contributing to the advancement of translational and clinical research.
How does it feel to win the 20th WRIB Poster award?
Winning the 20th WRIB Poster Award was a tremendous honor. I am incredibly grateful for the recognition and for the opportunity to share innovative science with such a respected scientific community. This achievement reflects the collaborative efforts of many talented colleagues, and I am especially thankful for their support throughout the project. The experience has been both rewarding and motivating, and I look forward to continuing to learn, grow, and contribute meaningful scientific work in the future.
Can you tell us a little about the work on which the Poster was based?
The poster focused on the development of a fit-for-purpose multiplex hybrid LC-MS/MS workflow that combines online and offline anti-peptide immunoprecipitation techniques. This approach enables the sensitive and selective quantitation of protein biomarkers and binding partners from a single digested heart tissue biopsy sample across multiple species using a validated, ICH M10-aligned methodology. By integrating these technologies, we developed a novel workflow capable of quantifying multiple protein biomarkers from a single tissue sample while minimizing sample processing requirements. What were the key conclusions from your research?
This work demonstrated a Context of Use (CoU) GLP validation strategy that leverages a single sample from a rare matrix to monitor multiple biomarkers using an advanced online antibody column and an offline bead-based hybrid LC-MS/MS platform simultaneously. The approach significantly reduces sample consumption while improving efficiency through shorter turnaround times. These advantages make it a valuable approach for supporting translational research and biomarker-driven drug development, particularly when only limited amounts of precious samples, such as biopsy tissue, are available.
What are you looking forward to working on over the next year?
Over the next year, I am excited to continue contributing to innovative scientific research and advancing novel analytical approaches. From a scientific perspective, I am particularly excited about supporting emerging therapeutic modalities, including antibody-oligonucleotide conjugates (AOCs), bispecific antibodies, and dual-conjugate antibody-drug conjugates (ADCs), as well as advancing biomarker strategies across multiple mass spectrometry platforms, including triple quadrupole (QqQ) and high-resolution mass spectrometry (HRMS). Hybrid LC-MS technologies play an increasingly important role in these areas, and I look forward to contributing to their development and application. I also look forward to expanding my interactions with clients, strengthening collaborative partnerships, and gaining broader experience in project leadership. Additionally, I am eager to continue developing toward future Principal Investigator (PI) responsibilities while contributing to the success of both our clients and KCAS Bio.

With more than 1,200 attendees and a highly selective review process, Workshop on Recent Issues in Bioanalysis (WRIB) represents a leading forum for excellence in bioanalytical science. This year, over 50% of submitted abstracts were not accepted, underscoring the rigor of the selection process. Poster presentations are evaluated based on scientific innovation, relevance to the field, and overall research quality.
